Synthetic phosphopeptide derived from a region of human Vav1 that contains Tyr160.
Vav proteins belong to the guanidine nucleotide exchange factor family of proteins that couple receptors to Rho-GTPases. The Vav family has currently three members in mammalian cells (Vav1, Vav2, and Vav3). Vav1 contains multiple protein binding motifs, indicating a role for Vav1as an adaptor protein linking cell surface receptors to downstream signaling proteins. Vav1 is exclusively expressed in hematopoietic cells and plays a significant role in lymphocyte development and antigen receptor mediated activation of NFAT and NFkB, and in more delayed responses such as the production of cytokines, including IL-2, IL-3, IFN-y, and TNF-a. Vav1 is also reported to activate JNK, p21-activated kinase (PAK) and PLCy, implicating its critical role in calcium mobilization, and cytoskeletal rearrangement. Activation of several cell surface receptors including integrins, immune response receptors, or growth factor receptors leads to phosphorylation of Vav1 on tyrosine residues including tyrosine 160. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Positive Control: K562 cells expressing wild-type human Integrin av and b3 proteins Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
