Vimentin (57kD) is the intermediate filament protein (IFP) of mesenchymal cells. It is the only IFP, which deviates from the tissue-specific and developmentally regulated pattern of expression. Besides its typical expression in most cultured cells, vimentin is also expressed together with several other IFPs during early stages of development. As differentiation proceeds, vimentin is exchanged for the tissue-specific intermediate filament type. Desmin (53kD) exhibits a high degree of tissue specificity, its expression being predominantly confined to all types of muscle cells (cardiac, skeletal and smooth muscle). Regulation of desmin expression is stage and tissue-specific, since it is induced during terminal development. In skeletal en cardiac muscle cells desmin is localized in the Z-disk region and at the intercalated disk. The expression pattern of desmin in smooth muscle is much more heterogenous. Coexpression of vimentin and desmin has been observed in rhabdomyosarcomas and leiomyosarcomas. Applications: Suitable for use in Immunohistochemistry, Flow Cytometry, Immunocytochemistry and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50-1:100 Immunohistochemistry (Frozen): 1:50-1:100 using ABC as detection reagent. Western Blot: 1:50-1:500 Optimum dilutions to be determined by researcher. Hybridoma: Sp2/0-Ag14 mouse myeloma cells with spleen cells of Balb/c mice. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
