Recombinant protein corresponding to Ser2-Glu466 from human Vimentin, expressed in E. coli.
Vimentin belongs to the intermediate filament family. It is highly expressed in mesenchymal cells and has been used as a specific mesenchymal marker. Vimentin has a role in cytoskeletal organization and cell morphology. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 cells. For intracellular staining, cells must first be fixed and permeabilized. The use of 4% PFA as a fixative and a 0.1% saponin balanced salt solution for permeabilization and washing is recommended. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Cells were harvested and washed twice in saline buffer. 2. Cell surface staining may be done at this point following the manufacturers staining procedure. 3. Up to 1x10e6 cells were resuspended in 500ul of cold 4% Flow cytometry fixation buffer and incubated at RT for 10 minutes. 4. Following fixation, the cells were washed twice in saline buffer, then once in saponin-containing saline buffer. 5. After permeabilization, 10ul of V2122-12A was added and the cells were incubated for 30 minutes at RT in the dark. 6. The cells were washed twice with saponin-containing saline buffer 7. The cells were resuspended in saline buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with PE-labeled rat IgG2A antibody. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
