Vaccinia Virus VH1-related phosphatase (VHR), also known as dual-specificity phosphatase 3 (DUSP3), dephosphorylates a variety of substrates at serine, threonine, and tyrosine residues. VHR has been shown to dephosphorylate the mitogen-activated protein kinases ERK1, ERK2, and JNK, but not p38. Phosphorylation of VHR at Y138 by ZAP70 enhances the inhibition of ERK2, suggesting a role for VHR in modulating T-cell receptor signaling. Applications: Suitable for use in Immunohistochemistry and ELISA. Other applications not tested. Recommended Dilutions: Immunohistochemistry: 25ug/mL to detect VHR in paraffin-embedded human tonsil tissue sections. ELISA (Direct): 0.5-1ug/ml.The detection limit for rhVHR is ~1ng/well. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
