Expression of p21WAF1 is induced by wild type, but not mutant, p53 suppressor protein. The p21WAF1 protein binds to cyclin/CDK complexes and inhibits their kinase activity thereby stopping cell cycle progression. Meanwhile, the p21WAF1 protein also binds to PCNA (proliferating cell nuclear antigen) and blocks DNA replication but not the DNA repair process. Evidently, inactivation of p53 disrupts the p53-dependent expression of p21WAF1. p21WAF1is a tumor suppressor involved in the pathogenesis of a variety of malignancies. Pooled monoclonal mouse IgGs Applications: Suitable for use in Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilution: Immunoprecipitation: 4ug immunoprecipitates Cip1 from 500ug of HeLa nuclear extract. Western Blot: 0.5-2ug/ml detects WAF1, p21 in 20ug of HeLa nuclear extract. HeLa nuclear extract was resolved by SDS PAGE, transferred to nitrocellulose, and probed with anti-WAF1, p21 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Positive Control: HeLa nuclear extract. Use 20ug per lane for minigels. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
