Human WISP1(Wnt-induced secreted protein1; also CNN4) is a 40kD, secreted, heparin-binding glycoprotein that is a member of the CCN (or CTGF/Cyr61/Nov) cysteine rich protein family. It is synthesized as a 367aa precursor that contains a series of sructural homology modules. Following a 22aa signal sequence, there is a 68aa IGFBP-like domain (aa53-120), a 57aa von Willebrand factor type C (VWC) module (aa126-182),a 40aa TSP type I domain (aa220-259) and a 75aa, C-terminal cysteine knot motif (aa273-347).The VWC module is associated with protein interaction, the TSP domain binds sulfated glycoconjugates, and the cysteine knot mediates dimerization and receptor binding. It is likely that WISP1 normally circulates as an 80kD homodimer. At least five alternative splice forms are known for WISP1. One is 30kD in size, 258aa in length, and shows a substitution of a His for (aa95-182). This removes the VWC domain. A second isoform is155aa in length and shows a frameshift at Arg 117 with a unique 38aa C terminal extension. A third is 195aa in length and shows a 31aa substitution for the first 203aa of the full length precursor. This retains the VWC and cysteine knot domains. A fourth shows a 43aa substitution for (aa117-367) for a total length of 163aa. This effectively removes everything but the IGFBPlike domain. The last splice form contains a deletion of (aa25-269) for a total length of 122aa. Thus, only the signal sequence and cysteine knot motifs are retained. This leaves only the IGFBPlike domain. Fulllength mature human WISP1 is 85% aa identical to both mouse and rat WISP1. WISP1 is expressed by osteoblasts and may contribute to fracture healing by promoting bone cell formation. Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilutions: ELISA: 2-8ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
