WWOX (WW domain-containing oxidoreductase, also WOX1 and FOR) is a 46kD cytoplasmic member of the short-chain dehydrogenases/reductases (or SDR) family of molecules. It is expressed in divergent cell types such as breast epithelium, keratinocytes, osteoblasts, and osteoclasts. WWOX has multiple effects, one of which is that of tumor suppressor. Here, WWOX is likely activated by either TGF-b binding to surface hyaluronoglucosaminidase 2, or C1q binding to C1qR, which, in both cases, initiates cell apoptosis. Human WWOX is 414aa in length and contains two WW (TrpTrp) domains (aa16-90) with an intervening NLS (aa50-55), plus an SDR region that contains a mitochondrial targeting sequence (aa121-330). There are multiple phosphorylation sites and multiple splice variants that impact intracellular localization. One shows a deletion of (aa173-352), a second shows a Lys substitution for (aa36-414), a third contains a 17aa substitution for (aa173-414, a fourth possesses a 76aa substitution for (aa138-414), a fifth shows an 11aa substitution for (aa353-414), while a sixth contains a 175aa substitution for (aa137-414). Over (aa36-126), human and mouse WWOX are identical in aa sequence. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
