X-Ray Repair Cross Complementing (XRCC1) plays a role in excision repair of DNA after ionizing irradiation. XRCC1 binds to DNA ligase III through the C-terminal 96 amino acids and to DNA polymerase beta through the N-terminal half.. In testis XRCC1 is expressed at high levels. Cells with a mutation of this gene exhibit decreased single strand break repair and reduced recombination repair. They show increased double strand breaks, and sister chromatid exchange is increased up to 10-fold. The XRCC1 might serve as a scaffold protein during base excision-repair. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: An initial concentration of 1-5ug/ml is recommended for Western. The antibody recognizes >70kD in HeLa cells and testis. Immunoprecipitation: 5-10ug/mg of protein lysate:. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
