Synthetic peptide corresponding to residues 282-307 of human Zap-70, following the second SH2 domain and preceding the kinase domain. (KLH coupled)
Applications: Suitable for use in Western Blot, Immunocytochemistry and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunocytochemistry: Detects Zap-70 in Jurkat cells. Western Blot: 0.5-1ug/ml Detects Zap-70 in RIPA lysates from Jurkat cells. Jurkat cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with Z0300-03D (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitates Zap-70 from 500ug of Jurkat RIPA lysate. Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Included Positive Antigen Control: J9000: Jurkat lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
