ZAP70, a 70kD member of the Syk tyrosine kinase family, plays a central role in lymphocyte activation and development, and is implicated in several immune disorders. Mutations in ZAP70 gene results in a form of Severe Combined Immunodeficiency Syndrome (SCID) in humans. ZAP70 expression also defines a subset of Chronic Lymphocytic Leukemia (CLL) in patients with unmutated Ig gene and poor clinical course. Applications: Suitable for use in Immunocytochemistry, Flow Cytometry and Western Blot. Other applications not tested. Recommended Dilution: Immunocytochemistry: 5-20ug/ml detected ZAP-70 in Jurkat cells fixed with formaldehyde and permeabilized with saponin. Western Blot: 1:500 detected ZAP 70 on 10ug of Jurkat lysates Optimal dilutions to be determined by the researcher. Positive Control: Western Blot: Jurkat cell lysate. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
