The principle is that PEPCK catalyzes the oxidation of oxaloacetate to produce phosphoenolpyruvate and CO2, pyruvate kinase and lactate dehydrogenase further catalyze the oxidation of NADH to produce NAD+ in turn, The rate of NADH decline was measured at 340 nm, which reflects PEPCK activity.
