3'-(O-Propargyl)-GTP can be incorporated in RNA at the 3end using poly(A) Polymerase. But for adding an alkyne to the 3'-end 3'-(O-Propargyl)-ATP (BCT-31) is superior. We developed a different approach in which 3'-(O-Propargyl)-GTP is added to mRNA to the 5end with an capping enzyme and afterwards methylated. As a result a cap-like structure is created which provides all the advantages of a cap structure together with the possibility to perform click reactions at the incorporated alkyne.
