ADS032 is a dual NLRP1/NLRP3 inhibitor that rapidly and reversibly blocks inflammasome formation, suppressing IL-1beta and TNF-alpha production in human immune and epithelial cells. It reduces NLRP3-driven ASC speck formation, protects mice from lethal influenza A by dampening lung inflammation, and inhibits Nigericin-induced IL-1beta secretion (IC50 = 94.6 µM, no washout, 354 µM, washout).
