P053 is a selective inhibitor of protein phosphatase 5 (PP5) - IC50 = 706 nM.1 PP5 regulates signaling cascades that are responsible for tumor initiation, progression, and metastasis and is thus an interesting anticancer target. P053 induced apoptosis in 786-O renal carcinoma cells. Inhibition of PP5 by P053 prevents deactivation of the death effector protein FADD and activates the extrinsic apoptosis pathway complex II.
