Each Path kit is specialized for the detection of an antigen-specific response. This unique combination of antigen with immunoassay is validated by scientists at Mabtech.

The human IFN-γ/IL-2 SARS-CoV-2 FluoroSpot kit enables enumeration of T cells secreting either one or both analytes in response to SARS-CoV-2 peptides. The combination of IFN-γ and IL-2 enables the quantitation of polyfunctional specific T cells.

The kit includes one low fluorescent plate pre-coated with monoclonal capture antibodies, SARS-CoV-2 S2 N defined peptide pool, SARS-CoV-2 SNMO defined peptide pool, and SARS-CoV-2 S1 scanning pool, monoclonal detection antibodies, secondary reagents conjugated to fluorophores, a fluorescence enhancer and an anti-CD28 monoclonal antibody for co-stimulation. A polyclonal activator for human T cells (mAb CD3-2) is supplied as a positive control for cytokine secretion. Pre-coated plates reduce assay time and minimize variability.

The SARS-CoV-2 S1 scanning pool contains 166 peptides from the human SARS-CoV-2 virus. The peptides are 15-mers overlapping with 11 amino acids, covering the S1 domain of the spike protein (amino acid 13-685). The pool is supplied in two vials: pool 1 SARS-CoV-2 S1 peptides 1-83 (83 peptides) and pool 2 SARS-CoV-2 S1 peptides 84-166 (83 peptides).

The SARS-CoV-2 S2 N defined peptide pool contains 41 synthetic peptides from the human SARS-CoV-2 virus. The peptides are derived from the spike and nucleoprotein.

The SARS-CoV-2 SNMO defined peptide pool contains 47 synthetic peptides from the human SARS-CoV-2 virus. The peptides are derived from the spike, nucleoprotein, membrane protein, ORF3a and ORF7a.

For analysis, we recommend the use of an automated FluoroSpot reader equipped with filters for excitation 490 nm/emission 510 nm (FITC) for 490-reagent detection and excitation 550 nm/emission 570 nm (Cy3) for 550-reagent detection. Filters should have high specificity to avoid bleed-through artifacts. Double secreting cells are identified by a computerized overlay.

IFN-γ
Interferon-γ (IFN-γ, also known as IFN-gamma, IFN-g, IFNg and Interferon-gamma) is produced mainly by activated T cells and NK cells. It is a proinflammatory cytokine that activates macrophages and endothelial cells, but it also regulates immune responses by effecting APC and T and B cells. Production of IFN-γ by helper T cells as well as cytotoxic T cells is a hallmark of the TH1-type phenotype, thus, high-level production of IFN-γ is typically associated with effective host defense against intracellular pathogens.

IL-2
Interleukin-2 (IL-2, IL2) is produced exclusively by activated T cells. IL-2 promotes the proliferation, differentiation, and survival of antigen-activated T cells and NK-cells.
