Human PDE4A1A (GenBank Accession No. U97584), full length protein with N-terminal GST tag (MW = 100kD), expressed in a Baculovirus infected Sf9 cell expression system. Specific Activity: Lot Specific. Sample Lot Value: 4,176 pmol/min/ug. Unit Definition: One unit is defined as the amount of enzyme that will convert 1 pmole of 3', 5'-cAMP to 5'-AMP per min at 37C. Assay Conditions: 10mM Tris-HCl, pH 7.4, 10mM MgCl2, 1mM MnCl2, 200uM cAMP, and 0.2ng/ul PDE4A1A at 37C for 20 min. Quantified by 5'-nucleotidase cleaving the 5'-AMP product and releasing the phosphate group which is detected by Malachite Green Reagent. Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
