A 50ul PKM2 reaction is conducted in a buffer containing 50mM Tris, pH 7.4, 100mM magnesium chloride, 250mM potassium chloride, 40mM ADP, 100mM phosphoenolpyruvate (PEP) at RT for 15 minutes. ATP production is detected using Kinase-Glo Luminescent Kinase Assay Platform. Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing. Store at -70C. Aliquots are stable for 6 months at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
