Vaccinia Virus VH1-related phosphatase (VHR), also known as dual-specificity phosphatase 3 (DUSP3), dephosphorylates a variety of substrates at serine, threonine, and tyrosine residues. VHR has been shown to dephosphorylate the mitogen-activated protein kinases ERK1, ERK2, and JNK, but not p38. Phosphorylation of VHR at Y138 by ZAP70 enhances the inhibition of ERK2, suggesting a role for VHR in modulating T-cell receptor signaling. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 0.5ug/ml with lysates of TE-85 human osteogenic sarcoma cell line, TF-1 human erythroleukemic cell line, and C2C12 mouse myoblast cell line. Conducted under reducing conditions. Optimal dilutions to be determined by the researcher. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
