The gammadelta T-cell receptor (TCR) is a heterodimer composed of gamma and delta chains, expressed on the T-cell surface and involved in antigen recognition. The engagement of the gammadelta TCR with antigen triggers the phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) within the CD3 chains. Unlike alphabeta T cells, human gammadelta T cells recognize and kill transformed cells in a manner unrestricted by human leukocyte antigen (HLA) presentation. gammadelta T lymphocytes primarily develop in the thymus, where their defining receptor is generated through RAG-mediated recombination of a relatively limited repertoire of variable (V), diversity (D), and joining (J) gene segments. In addition to the T-cell receptor, gammadelta T cells often express activating natural killer (NK) receptors, which bind to stress-induced surface molecules frequently expressed on malignant cells.

Flow cytometry:1X10 6 Human PBMC were surface-stained with ABflo 450 Mouse anti-Human CD3 mAb (A25189,5 µl/Test) and FITC Rabbit IgG isotype control (A25616,5 µl/Test,left) or FITC Rabbit anti-Human TCR gamma/delta mAb (A28422,5 µl/Test,right). Cells in the lymphocyte gate were used for analysis.
