CD244, also known as 2B4, is a type I transmembrane protein with a molecular weight of 38 kD. CD244 is one of the members of the CD2 subsett of the immunoglobulin superfamily (igSF) molecules. It is expressed in a wide variety of immune cells, including NK cells, most CD8+ T cells, most gamma/delta´ T cells, monocytes, eosinophils, and basophils. CD48 is the ligand of CD244. It has been reported that the ligation of human CD244 can cause enhanced NK cell cytotoxicity as well as cytokine production. Recent studies have indicated that human CD244 has inhibitory and activating functions, similar to that of murine CD244. These functions are dependent upon the density of 2B4 expression of the cell surface, the degree of ligation, and the level of adaptor molecule SAP expression.
