Source : E. coli,p53 is a DNA-binding protein that belongs to the p53 family. p53 is expressed ubiquitously and its isoforms are expressed widely in normal tissues but in a tissue-dependent manner. It is well known for its key role as a tumor suppressor protein. p53 induces growth arrest or apoptosis depending on the physiological circumstances and cell type. It is also involved in cell cycle regulation as a trans-activator that acts to negatively regulate cell division by controlling a set of genes required for this process. Whilst the activation of p53 often leads to apoptosis, p53 inactivation facilitates tumor progression. Mutants of p53, which frequently occur in different human cancers, fail to bind the consensus DNA binding site, and thus cause the loss of tumor suppressor activity. Defects in TP53 are a cause of esophageal cancer, Li-Fraumeni syndrome, lung cancer and adrenocortical carcinoma.
