TL1A, also known as TNFSF15, is a member of the TNF superfamily. This type II transmembrane protein forms stable homotrimers via its TNF homology domain (THD) and is expressed in immune cells, including monocytes, macrophages, dendritic cells, and T cells, as well as nonimmune cells like synovial fibroblasts and endothelial cells. By binding to Death Receptor 3 (DR3), TL1A modulates proliferation, activation, apoptosis, and cytokine/chemokine production in effector cells, contributing to both innate and adaptive immune homeostasis.
