Product Description

Alzheimers disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1, PSEN2) or in the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Several alternatively spliced transcript variants encoding different isoforms have been identified for this gene, the full-length nature of only some have been determined.

25 ug of the indicated Human whole cell extracts was loaded onto a 12% SDS-PAGE gel. 1 ug/mL of the antibody was used in this experiment.

The PSEN1 antibody (ARP58941) works fine in 4% paraformaldehyde fixed tissue sections (30 um), they were cut on a sliding, freezing microtome. We stained it overnight 1:500 followed with a biotinylated secondary, followed by an Avidin/peroxidase tertiary antibody, final staining with metal-intensified DAB. Similarly, both LRP antibodies (ARP58562 &ARP58563) work fine (stain appropriately) with this procedure. We are still evaluating the BACE1 antibody, it seems to work, too. -Dr. Thomas van Groen
