This kit detects AST/GOT activity by catalyzing the transamination of alpha-ketoglutarate and L-aspartic acid to produce pyruvate. The rate of absorbance increase at 505 nm is used to calculate AST/GOT activity.

In the assay, Aspartate aminotransferase (AST) catalyzes the transamination reaction of alpha-ketoglutarate and L-aspartic acid at 37C and pH 7.4 to produce glutamic acid and pyruvate, Pyruvate can interact with 2,4- Dinitrophenylhydrazine to produce pyruvate phenylhydrazone, which appears brownish red under alkaline conditions and has a characteristic absorption peak at 505 nm. The rate of pyruvate phenylhydrazone increase at 505 nm can reflect AST/GOT activity
