In the assay, NADK catalyzes the phosphorylation of NAD+ to produce NADP+, NADP+ can be reduced to NADPH by glucose-6-phosphate dehydrogenase. NADPH has a characteristic absorption peak at 340 nm. The rate of NADPH increase at 340 nm can reflect NADK activity
