It utilizes the red fluorescent dye sulforhodamine 101 (also known as Texas Red), conjugated to the target amino acid leucine and equipped with a chloromethyl ketone (CMK) reactive group, to create the fluorescent inhibitor probe SR101-Leu-CMK. SR101-FLISP specifically labels active chymotrypsin-like serine proteases in living cells.
