The Y187 strain is a commonly used strain in GAL4 system yeast hybrid experiments. It is of MATa type and can directly transform plasmids, and also can be used for library screening experiments with MATa type strains (Y2HGold, AH109, etc.) through mating. The screening markers are html, leu2, and the reporter genes are lacZ, MEL1. The Y187-pHis2 system yeast requires two plasmids, pHis2 and pGADT7Rec2. The screening marker for plasmid pHis2 is Trp, which is used to express pBait-His2 construct (cloned into pHis2 after repeated concatenation of 1-3 bait DNA sequences). The screening marker for plasmid pGADT7Rec2 is LEU, which is used to express the fusion protein of AD (GAL4 C-terminal 768-881 amino acids) and the target protein (Prey). The Principle for Y187-pHis2 Y1H is that: when the prey protein (Prey) binds to the bait sequence (BaitDNA), GAL4 AD activates the expression of HIS and can grow on a medium containing 3-AT, thereby determining the interaction between DNA and protein.
