The Y1H pHis2 system is a commonly used system for validation the interaction between promoters and transcription factors or screening upstream regulatory proteins of promoters. The vector usded for this Y1H is pHis2 and pGADT7, with host Y187. The pHis system can select a sequence (mainly concentrated region of elements) or a full-length, generally below 2000bp, and construct it on the pHIS2 vector. Due to the fact that a sequence contains more than one element, more interacting proteins are screened out or the probability of verifying interactions is higher, but it cannot be directly determined which specific element it interacts with. This kit contains the basic culture medium and reagents required for library screening by Y1H pHis2 system.
