The NMY51 yeast hybrid system is based on the DUAL Membrane system, with appropriate modifications by immobilizing cytoplasmic proteins (or nucleoproteins) onto the membrane through a small membrane protein (yeast ER protein Ost4) for easy detection. Firstly, the N-terminus of Bait protein (Bait-Cub-LexiA-VP16) is fused with a small membrane anchor protein (Ost4) to form a fusion protein (Ost4-Bait-Cub-LexiA-VP16). Then, the target protein is fused to the N-terminus of the ubiquitin NubG moiety. If there is an interaction between bait and prey, it promotes the proximity of the NubG and Cub of ubiquitin, resulting in the reconstruction of separated ubiquitin and recognition by UBPs, leading to the dissociation of LexA-VP16 into the nucleus and activation of reporter gene transcription.
