Cucurbitacin B is a natural product isolated from the Cucurbitaceae family.1 It displays a broad range of pharmacological activities including anti-inflammatory, antioxidant, antiviral, hypoglycemic, hepato-and neuroprotection, and anti-cancer.1,2 Cucurbitacin Bs activities are the result of regulation of diverse signaling pathways including JAK/STAT3, Nrf2/ARE, NFkB, AMPK, MAPK, and others. It has recently been shown to block the m6A RNA reader protein IGF2BP1 (IC50 = 4.1 µM and 1.0 µM 24 and 72 hr respectively).3 IGF2BP1 blockade in hepatocellular carcinoma cells leads to apoptosis and subsequent recruitment of immune cells to the tumor microenvironment and PD-L1 blockade.
