NK20a is a peptide designed from the structure of the complex of interleukin-10 (IL-10) with IL-10Ra, the ligand binding subunit of the IL-10 receptor. NK20a has excellent specific affinity for IL-10Ra and can function as an IL-10 antagonist. IL-10 is an immune regulatory cytokine with immunosuppressive and anti-inflammatory activity produced by various cells, including T cells, B cells, keratinocytes, mast cells, eosinophils, dendritic cells, monocytes, and macrophages, and it is upregulated in various cancers. In vitro, NK20a exerts anticancer effects on lymphoma B cells and can abolish the suppression effect of IL-10 on macrophages, consistent with the idea that blockade of the IL-10 receptor inhibits cell proliferation and survival. NK20a can also be conjugated with gold nanoparticles to enhance the anticancer efficacy of 5-fluorouracil (5-FU) against the breast cancer cell line BT-474.
