Protonectin-F is a modified peptide based on protonectin peptide from the venom of the wasp Parachartergus fraternus, which exhibits higher antinociceptive activity and less motor impairment than the parent peptide. The activity of protonectin-F is decreased by either of naloxone and AM251, antagonists of opioid and cannabinoid receptors respectively, which are two critical antinociception pathways. Protonectin-F has no activity on the acid-sensing ion channels (ASICs) rat ASIC1a and ASIC1b, human ASIC1a, and rat ASIC2a. In vivo, in a murine thermal pain induction model, protonectin-F had an antinociceptive effect comparable to morphine at same dose, while inducing less tolerance in after repeated administration.
