DPN is a potent estrogen ERbeta receptor agonist with 70-fold selectivity over ERalpha, at each of which it has EC50 values of 0.85 and 66 nM respectively. In vitro, DPN is able to relax mesenteric arteries and can regulate expression of GluR1, GluR2 and GluR3 in rat hippocampus. DPN stimulates proliferation of the androgen-independent prostate cancer cell line PC-3 via a novel pathway involving ERbeta-mediated activation of beta-catenin and promotes the migration and invasion of androgen€'independent DU€'145 prostate cancer cells. In vivo, DPN is able to ameliorate portal hypertension in a carbon tetrachloride induced liver cirrhosis rat model.
