PS18 is a peptide derived from the NH2-terminal hydrophilic sequence of the rat saposin C-domain. Saposin and its precursor prosaposin are endogenous proteins with neurotrophic and anti-apoptotic properties, and PS18 mimics the trophic activity of full length prosaposin. PS18 is neuroprotective in cellular and animal models of Parkinsons disease, and in a rat Alzheimer model, PS18 attenuates Abeta1-42-induced neuroinflammation and prevents memory deficits. In gerbils, PS18 protects against ischemia induced learning disability and neuronal loss, and improves behavioural deficits and enhances survival of neurons against neurotoxicity induced by kainic acid. The mechanism of action of PS18 is complex, it activates the PI3K/Akt pathway, phosphorylates GSK-3alpha/beta, suppresses the caspase pathway, upregulates Bcl-2 and downregulates BAX. The receptors for prosaposin are proposed to be GPR37 and GPR37L1.
