MG 132 is a specific inhibitor of the chymotrypsin-like activity of the 20S proteasome with an IC50 of 100 nM with Z-LLL-AMC as the substrate. MG 132 also inhibits calpain, suppresses gastric cancer cell proliferation and induces macro-autophagy. It also inhibits TNF-alpha-induced NF- kappa B activation and I kappa Balpha degradation and induces neurite outgrowth in PC12 cells. MG 132 inhibits SARS-CoV-2 Mpro in vitro and can inhibit viral replication.
