Ac-RLR-AMC (Ac-Arg-Leu-Arg-AMC) is a fluorogenic peptide substrate used to detect the trypsin-like peptidase activity of the 26S proteasome. Proteasome activity is measured using Ac-RLR-AMC by detecting the aminomethylcoumarin (AMC) released via hydrolysis. Ac-RLR-AMC serves as a substrate in proteolytic assays to analyze the regulation of 26S proteasome trypsin-like activity[1]. The maximum excitation wavelength and maximum emission wavelength of AMC are 380 nm and 440-460 nm, respectively.
