VCP Activator 1 is a valosin-containing protein (VCP/p97) activator with a Kd value of 27 µM. VCP Activator 1 binds to an allosteric pocket near the VCPC terminus in both the apo and ADP-bound states of VCP, mimics the interaction of the phenylalanine residue at the VCPC C-terminal tail, regulates enzymatic activity and stimulates ATP hydrolysis. VCP Activator 1 identifies a druggable allosteric site of VCP, provides a tool for investigating the enzymatic mechanism of action, and can be used to verify hypotheses associated with VCP loss-of-function protein aggregation diseases. VCP Activator 1 is applicable to the research of amyotrophic lateral sclerosis[1].
