AP-6 is a TMEM175 channel blocker with an IC50 value of 141 µM against human targets (under neutral pH conditions), and it exhibits selectivity over other 4-AP-sensitive potassium channels. AP-6 binds to the luminal entrance of the ion-permeable pathway of TMEM175, thereby blocking ion conduction and inhibiting the flow of potassium ions and protons through this channel.
