WX-02-43 is a stereoselective pan-FOXA ligand that covalently binds to specific cysteine residues of the spliceosomal component SF3B1 and the transcription factors FOXA1/2/3. By remodeling the chromatin-binding profile of FOXA1 and relaxing its DNA-binding preference, WX-02-43 induces bidirectional changes in chromatin accessibility and nucleosome invasion in prostate cancer cells, thereby stereoselectively regulating de novo gene expression. WX-02-43 exhibits significant SF3B1 binding-dependent antiproliferative activity in prostate cancer cells such as 22Rv1, but shows no obvious effect on most other human cancer cell lines. WX-02-43 is applicable for prostate cancer research[1][2][3].
