The SMAD3 protein acts as a receptor-regulated SMAD (R-SMAD), transducing signals from TGF-beta and activin type 1 receptor kinase. After being activated by antigen-presenting cells, SMAD3 binds to the TRE element in the gene promoter and forms a complex with SMAD4 to activate transcription.
