BTLA/CD272 protein inhibits antigen receptor signaling via PTPN6/SHP-1 and PTPN11/SHP-2. It regulates immune responses through cis and trans interactions with TNFRSF14. In cis, it maintains resting state in naive T cells, while in trans, it supports survival signals in effector T cells. BTLA/CD272 interacts with PTPN6/SHP-1, PTPN11/SHP-2, and TNFRSF14/HVEM.
