Lipid 114 is an ionizable cationic lipid with a pKa of approximately 6.8. It is utilized to create lipid nanoparticles (LNPs) for delivering siRNA both in vivo and in vitro. Lipid 114 LNPs encapsulating siRNA targeting IL-1beta can reduce IL-1beta expression in macrophages. Additionally, these LNPs lower IL-1beta expression in the liver and kidneys and alleviate liver inflammation in a mouse model of LPS-induced acute liver failure.
