The Carboxymethyl Lysine (CML) BioAssay(TM) ELISA Kit is a competitive inhibition immunoassay for the in vitro quantitative measurement of CML in serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids. Detection Range: 61.7-5000ng/ml Sensitivity: 25.9ng/ml Intra-Assay CV: <10% Inter-Assay CV: <12% Kit Components: *023914A: Microtiter Strips, 1x96 wells, Pre-coated, ready to use. *023914B: Standard, 2x1vial 023914C: Standard Diluent, 1x20ml *023914D: Detection Reagent A (green), 1x120ul *023914E: Detection Reagent B (red), 1x120ul 023914F: Assay Diluent A, 1x12ml 023914G: Assay Diluent B, 1x12ml 023914H: TMB Substrate, 1x9ml *023914I: Positive Control, 740.1pg/ml, 1x1vial (Lyophilized) *023914IJ: Stop Solution, 1x6ml 023914K: Wash Buffer, 30X, 1x20ml Precaution: The Stop Solution (023914IJ) included for use in this kit is an acid solution. Wear eye, hand, face, and clothing protection when using this material. Storage and Stability: Store *023914A, *023914B, *023914D, *023914E and *023914I at -20C. After reconstitution of store at 4C and use within 5 days. Store all the other components at 4C. Unused kit is stable for 6 months after receipt. Once kit components are opened, it is highly recommended to use remaining reagents within 1 month provided this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap. Usage: Reconstitute the standard with 0.7ml, of standard diluent, kept for 10 minutes at RT, shake gently (not to foam). The concentration of the standard in the stock solution is 5000ng/ml. Please prepare 5 tubes containing 0.6mL Standard diluent and produce a triple dilution series. Mix each tube thoroughly before the next transfer. Set up 5 points of diluted standard such as 5000ng/ml, 1666.7ng/ml, 555.6ng/ml, 185.2ng/ml, 61.7ng/ml, and the last EP tubes with standard diluent is the blank at 0ng/mL Assay Procedure Summary: 1. Prepare all reagents, samples and standards, 2. Add 50ul standard or sample to each well. 3. Add 50ul prepared Detection Reagent A immediately. Shake and mix. Incubate 1 hour at 37C. 4. Aspirate and wash 3 times. 5. Add 100ul prepared Detection Reagent B. Incubate 30 minutes at 37C. 6. Aspirate and wash 5 times. 7. Add 90ul Substrate Solution. Incubate 10-20 minutes at 37C. 8. Add 50ul Stop Solution. Read at 450nm immediately.
