ErbB4, also known as Her4, is a receptor tyrosine kinase that is a member of the epidermal growth factor receptor family. ErbB family members serve as receptors for the EGF family of growth factors. It is a single-pass type I transmembrane protein with multiple furin-like cysteine rich domains, a tyrosine kinase domain, a phosphotidylinositol-3 kinase binding site and a PDZ domain binding motif. It is expressed in normal skeletal muscle, heart, pituitary, brain and several breast carcinomas. ErbB4 ligands include the neuregulins, beta cellulin and heparin-binding EGF-like growth factor (HB-EGF). Monomeric ErbB4 binds its ligands with low affinity. ErbB4 appears to play important roles in neuronal development, development of the heart and cancer. Single nucleotide polymorphisms in ERBB4 have also been found in a study of patients with familial amyotrophic lateral sclerosis type 19. Recombinant human ErbB4, fused to hIgG-His-Tag at C-terminal, was expressed in HEK293 and purified by using conventional chromatography techniques.
