Hsp90bis a molecular chaperone with essential functions in maintaining transformation. Inhibition of Hsp90b functions has been shown to play a role in tumorigenesis and disease progression. The Hsp90b Assay Kit is designed for identification of Hsp90b inhibitors using fluorescence polarization. The assay is based on the competition of fluorescently labeled geldanamycin, an HSP90 inhibitor, for binding to purified recombinant Hsp90b. Intended Use: Designed for identification of Hsp90b inhibitors using fluorescence polarization. Test Principle: The Hsp90b inhibitor screening assay kit comes in a convenient 96-well or 384-well format, with purified Hsp90b enzyme, FITC-labeled geldanamycin, and Hsp90 assay buffer for 100 enzyme reactions. The key to the Hsp90b Assay Kit is the fluorescently labeled geldanamycin. Using this kit, only one simple step on a microtiter plate is required for Hsp90b reactions. The FITC-labeled geldanamycin is incubated with a sample containing Hsp90b enzyme to produce a change in fluorescent polarization that can then be measured using a fluorescence reader. Kit Components: Hsp90b recombinant enzyme: 1x70ug FITC-labeled geldanamycin (2.5uM): 1x30ul Hsp90 assay buffer (5X): 1x4ml Black, low binding NUNC microtiter plate: 1 plate Storage and Stability: Store other components at 4C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
