A cell-permeable aminopyridine acetamide based compound that acts as an inhibitor of SH2 domain-containing inositol 5-phosphatase 2 (SHIP2) that can overcome insulin resistance in 3T3-L1 adipocytes. Enhances insulin-stimulated Akt phosphorylation (Thr308) in TNFa-treated 3T3-L1 adipocytes (~10uM) and in primary cultured neurons from rat cerebral cortex. Improves glucose tolerance in db/db mice (~300mg/kg b.i.d) without affecting their fasting glucose levels. Also reduces the expression levels of glucose-6-phosphatase and phosphoenolpyruvate carboxykinase.
