Caged GTP is a fluorescent analog of guanosine triphosphate (GTP) that can be used to monitor the activity of the protein kinase, PKC. It is a caged compound that is hydrolyzed by esterases or glucuronidases in cells. This activation process releases free GTP and allows it to bind to its target protein. Caged GTP has been used in studies on ventricular myocytes and fetal bovine heart tissue, where it has been shown to inhibit protein synthesis by cross-linking with ribosomes. This inhibition may be due to its ability to generate carbonyl groups that are able to inhibit the enzyme ribonucleotide reductase, which is needed for RNA synthesis. The use of caged GTP has also been shown in a model system using wild-type mice and cross-linking agents such as forskolin.
