Ribavirin is a nucleoside analog that inhibits the replication of both DNA and RNA viruses, including influenza type A and type B. Ribavirin, as ribavirin 5-phosphate, affects virus-infected cells primarily by inhibiting the enzyme inosine 5-monophosphate (IMP) dehydrogenase. It potently suppresses eIF4E-mediated oncogenic transformation of murine cells in vitro, of tumor growth of a mouse model of eIF4E-dependent human squamous cell carcinoma in vivo, and of colony formation of eIF4E-dependent acute myelogenous leukemia cells derived from human patients. Ribavirin is a so-called antimetabolite, i.e. a drug that is chemically similar to naturally occurring metabolites but interfering with normal metabolic pathways. It is an antiviral nucleoside analog to the natural ribonucleoside guanosine (1). Ribavirin is a broad-spectrum antiviral drug, belonging to the group of inosine monophosphate (IMP) dehydrogenase inhibitors (2). Acting as a virostatic agent, ribavirin is used as an effective drug against respiratory syncytial virus (RSV), herpes viruses, influenza viruses type A and B, Hantaan virus, hepatitis C virus, adenoviruses, flavi viruses, reo viruses, and others.
