The protein encoded by this gene belongs to a family of phosphatidylinositol kinase-related kinases. These kinases mediate cellular responses to stresses such as DNA damage and nutrient deprivation. This protein acts as the target for the cell-cycle arrest and immunosuppressive effects of the FKBP12-rapamycin complex. The ANGPTL7 gene is located in an intron of this gene. [provided by RefSeq, Sep 2008]. Source: Recombinant protein corresponding to human MTOR/Raptor/MLST8 Complex, fused to FLAG-tag (mTOR) and His-tag (Raptor/MLST8) at N-terminal, expressed in Sf9 insect cells. Molecular Weight: ~324kD (complex) Specific Activity: 7.1pmol/min/ug Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Assay Conditions: Enzyme reaction is conducted in a buffer containing 50mM HEPES, pH 7.5, 10mM MgCl2, 1mM EGTA, 200um ATP, 0.01% Brij-35 and 2um substrate (Zlyte Ser/Thr peptide 11) at RT for 1 hour. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
