Secretogranin-5 is an acidic protein of 186aa and has an expression that seems to be restricted to neurons and endocrine cells. In the brain, it is most abundant in the supraoptic and paraventricular nuclei of the hypothalamus. It acts in neuroendocrine cells as a specific chaperone for proprotein convertase-2 (PC2) which is an important endoprotease involved in the processing of neuroendocrine precursors. It binds to inactive proPC2 in the ER and facilitates its movement toward the secretory pathway where proPC2 is proteolytically matured and activated. It also plays a role in regulating pituitary hormone secretion.
